I think there are two situations regarding your problem. First, your Spybot installation might have been corrupted. Second, a malware infection has interfered with your Spybot’s function. What you should do is this; Download and Install a Super AntipSpyware. Perform a full and thorough full system scan in safe mode. You might also want to Perform Spybot Search and Destroy scan. Uninstall your old Spybot S&D and install a new one. Check this. The home of Spybot-S&D!.

The tendency of malware is the ability to “lock” a used file in normal Windows mode. This means, your Spybot doesn’t function properly because of malware infection while in normal mode. It has issues with an infected Zlob file because it’s running and “locked” which clearly tells why your Spybot S&D cannot finish its function. But, under a safe mode, removal tools like I have mentioned earlier can detect malware or spyware better because these programs are not running. Ergo, it cannot lock or infect a running file. Once malware or spyware are detected, SuperAntiSpyware and Spybot Search and Destroy can then remove them easily.
